PRINCETON, N.J., Nov. 25 /PRNewswire-FirstCall/ -- Orchid BioSciences, Inc. (Nasdaq: ORCH - News) today announced that its Orchid Cellmark unit was awarded two contracts from the U.S. Federal Bureau of Investigation to develop specific aspects of the Company's SNP technology for additional forensic DNA testing applications. The goal of the two contracts is to develop new forensic tools that will increase the ability of investigators to identify individuals using DNA samples that have degraded and therefore contain a limited amount of genetic material. Work on both of these contracts is being undertaken in Orchid Cellmark's Dallas facility and is expected to be completed in about one year.

A SNP, or single nucleotide polymorphism, represents a single base difference in the more than three billion bases of DNA comprising the human genome and is the most common form of genetic variation. Since each person has a distinctive set of SNPs, SNP panels can be useful for identifying individuals using degraded or damaged DNA primarily because they can generate definitive matches using much shorter segments of DNA than other approaches.

The first contract is to develop an informative panel of SNP markers that can identify male DNA by measuring polymorphisms on the Y chromosome, the chromosome found only in men. This panel is designed to generate a profile of human male DNA from degraded samples, and is expected to be particularly useful in constructing the male profile in samples that contain mixtures of DNA from both a man and a woman. The contract is titled "Identification and Typing of Y-SNPs in Forensically Relevant Populations."

The informative Y-SNP panel could also potentially be used as a screening tool to derive broad physical characteristics based on information about a suspect's ancestral origin. These physical characteristics could be useful in so-called "no-suspect" crimes where investigators have DNA evidence but no other clues to identify possible perpetrators. Following the development of the Y-SNP panel, Orchid believes it would be the first private forensic lab to offer a gender-specific SNP-based panel as part of its services.

The second contract is to develop an expanded panel of robust SNP markers that will nearly double the number of SNPs Orchid currently uses, thereby potentially increasing its ability to identify individuals using degraded DNA. Titled "High-throughput Autosomal SNP Typing for Human Identification," this effort will build on the assay Orchid previously developed to help identify World Trade Center victims.

"Orchid's pioneering SNP analysis technology has a number of important applications to improve human identity profiling, so it is fitting that Orchid Cellmark was selected to develop these new tests by the FBI, the nation's top crime-fighting institution," said Mark D. Stolorow, executive director of Orchid Cellmark. "As a leading innovator in forensic genetic testing for nearly two decades, we look forward to further building on Orchid's proprietary SNP technology to expand our service offerings in order to better serve our law enforcement clients."

Orchid Cellmark has a long history of using advanced DNA analysis techniques for identity testing. It was among the first laboratories to use new DNA technology to identify casualties in the 1991 Persian Gulf War. Orchid's pioneering SNP technology is currently being used in collaboration with the New York Office of Medical Examiner in an attempt to identify the large number of 9/11 World Trade Center victims for whom conventional identification methods have failed.

The FBI is the investigative arm of the U.S. Department of Justice. The mission of the FBI is to protect and defend the U.S. against terrorist and foreign intelligence threats, to uphold and enforce the criminal laws of the U.S., and to provide leadership and criminal justice services to federal, state, municipal and international agencies and partners. More information on the FBI can be found at www.fbi.gov.

Police reopen PC Blakelock murder case
BY PA NEWS

Police have launched a fresh investigation into the murder of a police constable during riots in North London more than 18 years ago.

PC Keith Blakelock was hacked to death during the Broadwater Farm riots in Tottenham in 1985.

Winston Silcott was convicted in 1987 of his murder but the conviction was later overturned on appeal because of "unsafe" police evidence. He was freed from prison in October after serving 17 years for the murder of another man.

Scotland Yard today said that the Metropolitan Police Specialist Crime Directorate had now launched a new investigation into the murder.

Police are also reopening the case of the attempted murder of PC Richard Coombes, who was injured in the riots, a spokeswoman for Scotland Yard said.

The cases relating to both men had been under lengthy review since January 2000. Police said that the review had identified "new lines of inquiry" which were to form the basis of the new investigation.

The BBC reported that it believed DNA tests could be carried out on some exhibits in the case and that people were going to be re-interviewed.

Assistant Commissioner Tarique Ghaffur, head of the Specialist Crime Directorate, said that it would be a "tightly focused inquiry".

"The review took us in a number of directions, but there are some specific leads which we are now looking at," he said.

"We cannot go into exact details as this could seriously prejudice our inquiries. Suffice to say that we believe this information is strong enough to merit further investigation.

He said that the decision to reopen the case had been taken only after consultation with PC Blakelock's family.

"We accept that the decision to launch this fresh investigation will be difficult for some sections of the community," he said.

"MPS officers have consulted with a wide range of people in reaching this decision to reopen the case.

"We are grateful for the input we have received, particularly from Keith's family, the local community and the wider black community, whose views have greatly shaped how we will approach this new inquiry."

PC Blakelock, who left three sons, was set upon by a mob and hacked to death during the riots on October 6, 1985. The riots were sparked by the death of 49-year-old Cynthia Jarrett during a police raid. She died of heart failure after four police officers burst into her home.

Winston Silcott, Mark Braithwaite and Engin Raghip were all originally found guilty of PC Blakelock's murder, but the Court of Appeal overturned their convictions in 1991.

Less than two months ago, PC Blakelock's widow said that she still believed his killer would be brought to justice.

Elizabeth Johnson, 52, who now lives in Boldon Colliery, on the outskirts of Sunderland, Tyne and Wear, has since remarried.

She spoke after it emerged that Mr Silcott was to be freed from jail.

Mrs Johnson said at the time: "If the man is going to be let out of prison then nothing I can do will change that. But I do believe that guilty people will be brought to justice and we will see people being punished for what they did to Keith.

"They have deprived a family of a father and a grandfather. It never leaves you and it never goes away, you just get better at handling the situation. People cannot get away with murdering people and I believe that life should mean life. If they don't get what they deserve in this life they will in another."

DNA tests used in 1982 murder hunt

Miss Yianni was seen talking to a young man on her doorstep
Detectives are using new DNA forensic techniques to try to solve a murder in north London 21 years ago.
Yiannoulla Yianni, 17, was strangled and sexually assaulted at her home in Belsize Road, Hampstead in August 1982.

More than 1,000 people gave statements at the time and police now want all of them to come forward again.

They are offering a £10,000 reward for information which helps them convict Miss Yianni's killer, whom detectives believe she knew.

Postmen are delivering 5,000 leaflets to homes in and around Hampstead as part of the appeal.

Police say newly-developed forensic techniques have opened up new lines of inquiry and they are trying to trace everyone who gave statements or fingerprints at the time.

There must be people out there who know what happened

Detective Chief Inspector Richard Freeman
Detective Chief Inspector Richard Freeman said: "New advances in DNA techniques mean now we really can solve this case, but need the public to come forward and offer their help.

"This family have had to live with this tragic loss for over 21 years.

"There must be people out there who know what happened on that day and were reluctant to speak to police at the time.

"But 21 years on, whatever held them back may no longer apply."

Miss Yianni, who was Greek-Cypriot, spent the morning of 13 August 1982 at home with her mother.

They walked to the family shop in Fairhazel Road at 1230 BST and Miss Yianni went home alone to prepare dinner.

Her parents found her body when they returned home at 1500 BST.

Miss Yianni was seen talking to a man in his 20s, who was possibly Greek or Cypriot, on the doorstep of her home and police are still trying to find him.

They also want to hear from any of her friends from Quintin Kynaston School and colleagues from Woolworths in Finchley Road, both in St John's Wood.

They also want anyone who attended Greek dancing classes at 94 Camden Road, Camden, to come forward.
